Dr. Hope Rugo is Director, Breast Oncology Clinical Trials Program, University of California, San Francisco.

Low PTEN expression and PI3K mutations appear to predict the likelihood that HER2-positive breast tumors will be resistant to trastuzumab, while low PTEN predicts the likelihood of response to lapatinib, according to research reported at SABCS 2008. These findings suggest that the 2 anti-HER2 drugs act via different pathways and point to the possibility of more individualized therapy based on biomarker testing, says Dr. Hope Rugo.
