Bortezomib is currently the only single agent proven to prolong overall survival (OS) in patients with relapsed multiple myeloma (MM). The drug is active both as monotherapy and when given concomitantly with dexamethasone, doxorubicin (and pegylated liposomal doxorubicin), thalidomide, and other drugs. Rechallenge with bortezomib-containing regimens also appears be effective in patients with MM and is not associated with increased toxicity.
