In ovarian cancer, germline mutations of the BRCA1 and BRCA2 genes are associated with defective homologous recombination (HR), a key mechanism of repair of double-stranded DNA. Defective HR may be associated with sensitivity to platinum-containing chemotherapy. Impaired HR has also been shown to interrupt the base excision repair pathway, which is targeted by poly-ADP ribose polymerase (PARP) inhibition, a promising therapeutic mechanism for treating BRCA-mutant ovarian cancer.
